Natural Antibodies Produced in Vaccinated Patients and COVID-19 Convalescents Hydrolyze Recombinant RBD and Nucleocapsid (N) Proteins

Antibodies are protein molecules whose primary function is to recognize antigens. However, recent studies have demonstrated their ability to hydrolyze specific substrates, such as proteins, oligopeptides, and nucleic acids. In 2023, two separate teams of researchers demonstrated the proteolytic activity of natural plasma antibodies from COVID-19 convalescents. These antibodies were found to hydrolyze the S-protein and corresponding oligopeptides. Our study shows that for antibodies with affinity to recombinant structural proteins of the SARS-CoV-2: S-protein, its fragment RBD and N-protein can only hydrolyze the corresponding protein substrates and are not cross-reactive. By using strict criteria, we have confirmed that this proteolytic activity is an intrinsic property of antibodies and is not caused by impurities co-eluting with them. This discovery suggests that natural proteolytic antibodies that hydrolyze proteins of the SARS-CoV-2 virus may have a positive impact on disease pathogenesis. It is also possible for these antibodies to work in combination with other antibodies that bind specific epitopes to enhance the process of virus neutralization. Full article

Understanding the intricate relationships between diseases is critical for both prevention and recovery. However, there is a lack of suitable methodologies for exploring the precedence relationships within multiple censored time-to-event data, resulting in decreased analytical accuracy. This study introduces the Censored Event Precedence Analysis (CEPA), which is a nonparametric Bayesian approach suitable for understanding the precedence relationships in censored multivariate events. CEPA aims to analyze the precedence relationships between events to predict subsequent occurrences effectively. We applied CEPA to neonatal data from the National Health Insurance Service, identifying the precedence relationships among the seven most commonly diagnosed diseases categorized by the International Classification of Diseases. This analysis revealed a typical diagnostic sequence, starting with respiratory diseases, followed by skin, infectious, digestive, ear, eye, and injury-related diseases. Furthermore, simulation studies were conducted to demonstrate CEPA suitability for censored multivariate datasets compared to traditional models. The performance accuracy reached 76% for uniform distribution and 65% for exponential distribution, showing superior performance in all four tested environments. Therefore, the statistical approach based on CEPA enhances our understanding of disease interrelationships beyond competitive methodologies. By identifying disease precedence with CEPA, we can preempt subsequent disease occurrences and propose a healthcare system based on these relationships. Full article

Impact of Incorporating Future Mandatory Price Reductions with Generic Drug Entry on the Cost-Effectiveness of New Drugs: A Policy Simulation Study of Dupilumab in Atopic Dermatitis Treatment

The introduction of high-cost medications often poses challenges in achieving cost-effectiveness for drug insurance coverage. Incorporating future price reductions for these medications may enhance their cost-effectiveness. We examined the influence of future cost reductions mandated by the national insurer’s equal pricing for equivalent drugs (EPED) policy on the cost-effectiveness of dupilumab, a biologic drug for moderate to severe atopic dermatitis in the Korean healthcare system. We conducted a policy simulation study using semi-Markovian cost utility analysis of dupilumab in combination with supportive care (SC) versus SC alone, with and without the EPED policy adjustment. The EPED would lower dupilumab’s price to 70% following the entry of a biosimilar drug in 10.3 years. Scenario analyses quantified the impact of changing time to the EPED, chemical versus biological designation, response criteria, discount rates, and time horizons on the Incremental Cost-Effectiveness Ratio (ICER) and acceptability with and without EPED adjustment. The EPED adjustment of dupilumab’s future price significantly improved its cost-effectiveness, with a 9.7% decrease in ICER and a substantial 14.6% increase in acceptability. Assuming EPED in 5 years, the ICER fell below the predefined willingness-to-pay threshold. If dupilumab were a chemical drug, EPED adjustment demonstrated a 19.1% increase in acceptability. Incorporating future cost reductions via the EPED system in economic evaluations is crucial, especially for drugs facing imminent generic entry. This study underscores the importance of EPED adjustment in the cost-effectiveness analysis of innovative medications, especially for those nearing willingness-to-pay thresholds. Full article

Appraising the Physical Activity Levels of Saudis with Physical Disabilities: Effects of Disability Type, Mobility Assistive Devices, and Demographic Factors

Physical activity (PA) has numerous health benefits for individuals with physical disabilities (IWPD). However, it is common for activity levels to fall below the suggested limits. This study aimed to evaluate the prevalence, pattern, and levels of PA among IWPD in Saudi Arabia. It also investigated the effects of individuals’ type of disability, mobility assistive devices, and demographic features on PA levels. Data were collected from 238 participants, mostly male (62.2%), aged 39.76 ± 12.19 years. Among them, 19.3% had spinal conditions, 14.7% had progressive muscular dystrophy, 15.1% had multiple sclerosis, 17.6% had cerebral palsy, 16.4% had poliomyelitis, and 16.8% had limb or foot amputations. The participants were assessed using the Arabic version of the Physical Activity Scale for Individuals with Physical Disabilities (PASIPD-AR). The results showed that 62.6% (64.9% of males and 58.9% of females) met the minimum PA guidelines specified by the WHO. The average PASIPD-AR score was 10.33 ± 10.67 MET-hours/day, indicating lower PA levels, and 8.4% of individuals did not participate in any form of PA. Significant discrepancies were detected in disability type and mobility assistive device use after age adjustment. Marital status, education, and occupation greatly affected PA components. Greater attention should be paid to promoting an active lifestyle among IWPD in Saudi Arabia. Full article

Solid–liquid separation plays a decisive role in various industrial applications particularly in the treatment and purification of suspensions. Solid bowl centrifuges, such as the decanter centrifuge, are commonly employed in these processes as they operate continuously and enable high throughputs with short processing times. However, predicting the separation performance of solid bowl centrifuges proves to be challenging due to dynamic phenomena within the apparatus, such as particle settling, sediment build-up, consolidation and sediment transport. In practice, design considerations and the dimensioning of the apparatus rely on analytical models and the manufacturer’s expertise. Computational Fluid Dynamics (CFD) offers a way to deepen our understanding of these devices by allowing detailed examination of flow phenomena and their influence on the separation processes. This study utilizes the open-source software OpenFOAM to simulate multiphase flow in a laboratory-scale decanter centrifuge, solving individual transport equations for each particle size class. The basis is the characterization of the material through targeted laboratory experiments to derive material functions that describe the hindered settling and the sediment consolidation. Furthermore, experiments on a laboratory decanter served as validation. The results demonstrate the solver’s capability to replicate clarification and classification within the apparatus. Furthermore, the solver supports the definition of geometries tailored to specific separation tasks. This research demonstrates the potential of CFD for a better understanding of complex centrifuge processes and for optimizing their design to improve performance. Full article

Using an optimality theoretic analysis, this study offers a conception of the problem of plural realization in Eastern Andalusian Spanish (EAS) where plural suffix /s/ was deleted diachronically that differs from other accounts that assign the EAS plural an underlying suffixal /s/ synchronically. Using alignment constraints, we argue that plural /s/ does not appear in the underlying form synchronically in EAS, but that instead the plural morpheme is represented by a floating [–ATR]PL feature that aligns to the right edge of the word and spreads left. The [–ATR] feature, represented phonetically as a laxing or opening of vowels, applies to all mid vowels, low vowels in word final position, and combines with vowel epenthesis to explain Eastern Andalusian pluralization tendencies in words with final consonants. We discuss the behavior of high vowels, which can be transparent to harmony, and focus in particular on the plural of words that end in a final stressed vowel that have been rarely discussed in the EAS literature. We develop an optimality-theoretic analysis on the Granada variety and extend that analysis to other varieties with somewhat different patterns. Full article

Frailty is increasingly recognized as a significant health concern, particularly due to its association with cardiovascular pathologies. This study aims to examine how vascular endothelial dysfunction, a known premorbid stage in the pathophysiology of cardiovascular diseases, contributes to the link between cardiovascular illness and frailty. Methods: The inclusion criteria allowed us to focus on original clinical research articles published in English between January 2014 and January 2024, which reported quantitative assessments of the relationship between frailty and vascular endothelial dysfunction. Excluded from the study were systematic literature reviews, meta-analyses, editorials, conference articles, theses, methodological articles, and studies using animal or cell culture models. Searches were conducted of electronic databases, including Scopus, ScienceDirect, and Medline, up to 22 January 2024. The risk of bias was assessed using the Joanna Briggs Institute’s critical appraisal tools. The methods used to present and synthesize the results involved data extraction and categorization based on biomolecular and clinical findings of endothelial dysfunction. Results: Following the application of the inclusion and exclusion criteria, a total of 29 studies were identified. Vascular endothelial dysfunction was associated with increased frailty phenotypes, and we also identified SGLT-2 inhibitors’ potential role as an anti-fragility treatment that affects endothelial dysfunction. This study found that the physical and biomolecular markers of endothelial dysfunction are associated with frailty measures and have predictive value for incident frailty. Furthermore, some studies have shown inflammation to have an impact on endothelial dysfunction and frailty, and an innovative age-related chronic inflammation measure has been proven to predict frailty scores. Conclusions: The current evidence suggests an association between endothelial dysfunction and frailty, highlighting the need for further research to elucidate the underlying mechanisms. Full article

Recognizing the omnipresence of mathematics across various contexts, this paper addresses the untapped potential of museums as rich venues for informal mathematics learning beyond traditional educational settings like classrooms. This paper presents the InformalMath program, designed for the professional development of primary and middle school teachers using integrating mathematics education within art and history museums through designing mathematics visit workshops. Specifically, the focus is placed on Phase 1 of the program, in which teachers participated in two informal mathematics education workshops at two museums in Turin, Italy, and were asked to reflect on their participation through a written essay. The analysis of the essays reveals significant engagement, appreciation of mathematics as a cultural artifact, and the emergence of creativity and inclusion among participating teachers. These findings highlight the benefits of such interdisciplinary approaches in enhancing mathematical understanding and pedagogical strategies. Conclusions emphasize the program’s success in not only enriching teachers’ instructional repertoire but also in promoting a more holistic, engaging, and contextualized approach to mathematics education, suggesting a promising avenue for future educational practices and research in informal learning environments. A new approach for output feedback L1 adaptive control based on a proportional adaptation law is presented. The effectiveness of this design is assessed in simulation and validated through real-time testing on an airfoil pitch control wind tunnel experimental rig. Experimental evaluation of the robustness of the controllers, assessed by introducing various disturbances into the control signals, shows that the adaptive control has a better performance compared to PID control, particularly in scenarios with reduced control effectiveness and time-varying disturbances. The experimental results demonstrate the efficacy of the proposed method in practical applications. Full article

Plasmids are mobile genetic elements known to carry secondary metabolic genes that affect the fitness and survival of microbes in the environment. Well-studied cases of plasmid-encoded secondary metabolic genes in marine habitats include toxin/antitoxin and antibiotic biosynthesis/resistance genes. Here, we examine metagenome-assembled genomes (MAGs) from the permanently-stratified water column of the Cariaco Basin for integrated plasmids that encode biosynthetic gene clusters of secondary metabolites (smBGCs). We identify 16 plasmid-borne smBGCs in MAGs associated primarily with Planctomycetota and Pseudomonadota that encode terpene-synthesizing genes, and genes for production of ribosomal and non-ribosomal peptides. These identified genes encode for secondary metabolites that are mainly antimicrobial agents, and hence, their uptake via plasmids may increase the competitive advantage of those host taxa that acquire them. The ecological and evolutionary significance of smBGCs carried by prokaryotes in oxygen-depleted water columns is yet to be fully elucidated. Full article

Post-Conization HPV Vaccination and Its Impact on Viral Status: A Retrospective Cohort Study in Troms and Finnmark, 2022

Human papillomavirus (HPV) is associated with cellular changes in the cervix leading to cancer, which highlights the importance of vaccination in preventing HPV infections and subsequent cellular changes. Women undergoing the loop electrosurgical excision procedure (LEEP), a treatment for high-grade cervical intraepithelial neoplasia (CIN2+), remain at risk of recurrence. This study assessed the effect of post-conization HPV vaccination on the viral status of women at six months post-conization, aiming to evaluate the vaccine’s effectiveness in preventing recurrence of CIN2+. A retrospective cohort study was conducted among women in Troms and Finnmark who underwent conization in 2022. Using the SymPathy database and the national vaccination register (SYSVAK), we analyzed the vaccination statuses and HPV test results of women born before 1991, who had not received the HPV vaccine prior to conization. Out of 419 women undergoing conization, 243 met the inclusion criteria. A significant association was found between post-conization HPV vaccination and a negative HPV test at six months of follow-up (ARR = 12.1%, p = 0.039). Post-conization HPV vaccination significantly reduced the risk of a positive HPV test at the first follow-up, suggesting its potential in preventing the recurrence of high-grade cellular changes. However, the retrospective design and the insufficient control of confounding variables in this study underscore the need for further studies to confirm these findings. Full article

(1) Background: Uveal melanoma (UM) is a common malignant intraocular tumor that presents with significant genetic differences to cutaneous melanoma and has a high genetic burden in terms of prognosis. (2) Methods: A systematic literature search of several repositories on uveal melanoma diagnosis, prognosis, molecular analysis, and treatment was conducted. (3) Results: Recent genetic understanding of oncogene-initiation mutations in GNAQ, GNA11, PLCB4, and CYSLTR2 and secondary progression drivers of BAP1 inactivation and SF3B1 and EIF1AX mutations offers an appealing explanation to the high prognostic impact of adding genetic profiling to clinical UM classification. Genetic information could help better explain peculiarities in uveal melanoma, such as the low long-term survival despite effective primary tumor treatment, the overwhelming propensity to metastasize to the liver, and possibly therapeutic behaviors. (4) Conclusions: Understanding of uveal melanoma has improved step-by-step from histopathology to clinical classification to more recent genetic understanding of oncogenic initiation and progression. Abnormal Static Sagittal Cervical Curvatures following Motor Vehicle Collisions: A Retrospective Case Series of 41 Patients before and after a Crash Exposure

Previous investigations have found a correlation between abnormal curvatures and a variety of patient complaints such as cervical pain and disability. However, no study has shown that loss of the cervical curve is a direct result of exposure to a motor vehicle collision (MVC). This investigation presents a retrospective consecutive case series of patients with both a pre-injury cervical lateral radiograph (CLR) and a post-injury CLR after exposure to an MVC. Computer analysis of digitized vertebral body corners on CLRs was performed to investigate the possible alterations in the geometric alignment of the sagittal cervical curve. Methods: Three spine clinic records were reviewed over a 2-year period, looking for patients where both an initial lateral cervical X-ray and an examination were performed prior to the patient being exposed to a MVC; afterwards, an additional exam and radiographic analysis were obtained. A total of 41 patients met the inclusion criteria. Examination records of pain intensity on numerical pain rating scores (NPRS) and neck disability index (NDI), if available, were analyzed. The CLRs were digitized and modeled in the sagittal plane using curve fitting and the least squares error approach. Radiographic variables included total cervical curve (ARA C2–C7), Chamberlain’s line to horizontal (skull flexion), horizontal translation of C2 relative to C7, segmental translations (retrolisthesis and anterolisthesis), and circular modelling radii. Results: There were 15 males and 26 females with an age range of 8–65 years. Most participants were drivers (28) involved in rear-end impacts (30). The pre-injury NPRS was 2.7 while the post injury was 5.0; p < 0.001. The NDI was available on 24/41 (58.5%) patients and increased after the MVC from 15.7% to 32.8%, p < 0.001. An altered cervical curvature was identified following exposure to MVC, characterized by an increase in the mean radius of curvature (265.5 vs. 555.5, p < 0.001) and an approximate 8° reduction of lordosis from C2–C7; p < 0.001. The mid-cervical spine (C3–C5) showed the greatest curve reduction with an averaged localized mild kyphosis at these levels. Four participants (10%) developed segmental translations that were just below the threshold of instability, segmental translations < 3.5 mm. Conclusions: The post-exposure MVC cervical curvature was characterized by an increase in radius of curvature, an approximate 8° reduction in C2–C7 lordosis, a mild kyphosis of the mid-cervical spine, and a slight increase in anterior translation of C2–C7 sagittal balance. The modelling result indicates that the post-MVC cervical sagittal alignment approximates a second-order buckling alignment, indicating a significant alteration in curve geometry. Sepsis is one of the major causes of morbidity and mortality in hospitals, especially in low- and middle-income countries, and represents a challenge to health care providers to carry out early detection, and accurate diagnosis and prognosis with cost-effective diagnostic tools. An observational prospective study was conducted from December 2021 to December 2022 to investigate the extended inflammatory parameters (EIPs) for sepsis management and analyze the survival of septic patients in the emergency unit, intensive care unit (ICU) and inpatient ward. Patients suspected of having sepsis underwent a sequential organ failure assessment (SOFA) evaluation and had blood drawn for complete blood counts (CBCs). Significant changes were observed in various CBC parameters and EIPs, and the sepsis group was followed up with for 30-day mortality. The study highlighted a significant difference yet strong discriminatory power to differentiate sepsis with an AUC of 0.924 against the non-sepsis group and an AUC of 0.991 against the healthy control group using combination of white blood cells and EIPs. Furthermore, the study showed good predictive ability for 30-day mortality with a hazard ratio of 2.311. In summary, this study provides evidence that the utilization of EIPs may be valuable in diagnosing and predicting patient outcomes, and thus will be beneficial for sepsis management in the hospital. The aim of our study was to investigate iodine density (ID) and fat fraction (FF) on dual-energy computed tomography (DECT) in patients with acute pancreatitis (AP). This retrospective study included 72 patients with clinically confirmed AP and 62 control subjects with DECT of the abdomen. Two radiologists assessed necrosis and measured attenuation values, ID, and FF in three pancreatic segments. We used receiver operating characteristic (ROC) analysis to determine the optimal threshold for ID for the differentiation between AP groups. The ID was significantly higher in interstitial edematous AP compared to necrotizing AP and the control group (both p < 0.05). The ROC curve analysis revealed the thresholds of ID for detecting pancreatic necrosis ≤ 2.2, ≤2.3, and ≤2.4 mg/mL (AUC between 0.880 and 0.893, p > 0.05) for the head, body, and tail, respectively. The FF was significantly higher for pancreatitis groups when compared with the control group in the head and body segments (both p < 0.001). In the tail, the difference was significant in necrotizing AP (p = 0.028). The ID values were independent of attenuation values correlated with the FF values in pancreatic tissue. The growing increase in demand for water and the lack of balance between water supply and demand have led to conflicts among the downstream stakeholders of the international Hirmand River. This river is shared between Iran and Afghanistan and is located in the southeast of Iran, in the Sistan region. The Vardkhaneh is divided into two branches, Parian and Sistan, and it is the only main source of water in the Sistan region of Iran. The inner part of Hirmand catchment is considered bankrupt concerning its water resources, so there is a need to take the current status and resulting issues into account in order to resolve conflicts. In Hirmand catchment, four different games of bankruptcy theory, namely proportional (Pr), adjusted proportional (AP), constrained equal award (CEA), and constrained equal losses (CEL), were developed as optimization models based on genetic algorithms. For this purpose, the catchment was simulated with 26 scenarios using the WEAP 2022 version software for an average time period. The results of the bankruptcy game modeling showed that water could be effectively allocated to resolve conflicts among stakeholders. It is therefore recommended to use such a model to resolve fights and optimally allocate resources, even in bankrupt catchments. Full article

Currently, therapeutic and diagnostic applications of antibodies are primarily limited to cell surface-exposed and extracellular proteins. However, research has been conducted on cell-penetrating peptides (CPP), as well as cytosol-penetrating antibodies, to overcome these limitations. In this context, a heparin sulfate proteoglycan (HSPG)-binding antibody was serendipitously discovered, which eventually localizes to the cytosol of target cells. Functional characterization revealed that the tested antibody has beneficial cytosol-penetrating capabilities and can deliver cargo proteins (up to 70 kDa) to the cytosol. To achieve tumor-specific cell targeting and cargo delivery through conditional activation of the cell-penetrating antibody in the tumor microenvironment, a single-chain Fc fragment (scFv) and a VL domain were isolated as masking units. Several in vitro assays demonstrated that fusing the masking protein with a cleavable linker to the cell penetration antibody results in the inactivation of antibody cell binding and internalization. Removal of the mask via MMP-9 protease cleavage, a protease that is frequently overexpressed in the tumor microenvironment (TME), led to complete regeneration of binding and cytosol-penetrating capabilities. Masked and conditionally activated cytosol-penetrating antibodies have the potential to serve as a modular platform for delivering protein cargoes addressing intracellular targets in tumor cells. Cowpea is one of the most popular dry-land legumes cultivated for food and forage in arid and semi-arid areas. Genetic diversity for global germplasm can be organized into core collections providing optimum resources to serve breeding requirements. Here, we present diversity analysis and genome-wide association study (GWAS) results for part of the cowpea core collection of the United States Department of Agriculture (USDA) along with breeding line controls. Included in the analysis were a total of 373 accessions analyzed with 6880 Single Nucleotide Polymorphism (SNP) markers from Genotyping by Sequencing (GBS). Population structure differentiated accessions into two groups irrespective of geographical origin and formed three clusters based on taxa upon phylogenetic analysis. A total of 56 SNPs were significantly associated to nine traits including pod length (25 Quantitative Trait Nucleotides, QTNs), seed anti-oxidant content (7 QTNs), dry pod color (7 QTNs), plant maturity (5 QTNs), flower color (5 QTNs), seed weight (4 QTNs), tolerance to low phosphate (1 QTN), growth habit (1 QTN), and response to rock phosphate (1 QTN) using Bayesian-information, Linkage-disequilibrium Iteratively Nested Keyway (BLINK), and Fixed and random model Circulating Probability Unification (FarmCPU) association models. Key genes related to all significant SNPs were identified based on annotations of the cowpea reference genome, including a flavonoid gene controlling flower color (Vigun08g040200.1), a root nodulation regulator for tolerance to low phosphate (Vigun11g168000.1), and numerous genes involved in signaling, biosynthesis, metabolite transport, and abiotic stress. Our results highlight the importance of maintaining public phenotyping databases at USDA and strengthening collaborations for data collection in cowpea to maximize research impacts. Full article

Organic soil amendments are a sustainable option for modifying soil structure and improving plant performance in the face of abiotic stressors such as drought and soil salinity. Of these amendments, biochar and compost have the added benefits of carbon sequestration and waste recycling. Establishment studies were conducted on tall fescue (Festuca arundinacea Schreb.) (syn., Schedonorus arundinaceus (Schreb.) Dumort and Lolium arundinaceum (Scop.) Holub) to assess the potential inhibition of establishment by compost and biochar products. Both green waste and biosolid compost impaired establishment rates, while biochar did not. In the field study, the green waste treatments were slower to reach 50% coverage than the untreated control or when biochar was added to the soil, but all treatments reached 75% and final coverage at a similar rate. Field application of compost had a positive effect on final root length and volume but a negative effect on tall fescue roots in the greenhouse. The negative effect of higher salts and volatiles in the biosolids compost was reduced when biosolids and biochar were incorporated into the soil simultaneously. This work represents one of the only large-scale field studies on turfgrass establishment comparing the impact of biochar and compost products on turfgrass establishment. Full article

Heat Transfer Process of the Tea Plant under the Action of Air Disturbance Frost Protection

Wind machines based on the air disturbance method are progressively employed to mitigate frost damage within the agricultural machinery frost protection. These devices are utilized during radiative frost nights to disrupt near-surface thermal inversion through air mixing. Despite this application, the fundamental mechanisms underlying these mixing processes are not well comprehended. In this research, numerical simulations were conducted using COMSOL Multiphysics software version 6.0 to simulate the flow and heat transfer processes between the thermal airflow and both the tea canopy and stems. The results indicated that due to obstruction from the canopy cross-section, the airflow velocity on the contact surface rapidly increased. As the airflow further progressed, the high-speed region of the airflow gradually approached the canopy surface. Turbulent kinetic energy increased initially on the windward side of the canopy cross-section and near the top interface. On the windward side of the canopy, due to the initial impact of the thermal airflow, rapid heating occurred, resulting in a noticeable temperature difference between the windward and leeward sides within a short period. In the interaction between airflow and stems, with increasing airflow velocity, fluctuations and the shedding of wake occurred on the leeward side of the stems. The maximum sensible heat flux at the windward vertex of the stem increased significantly with airflow velocity. At an airflow velocity of 2.0 m/s, the maximum heat flux value was 2.37 times that of an airflow velocity of 1.0 m/s. This research utilized simulation methods to study the interaction between airflow and tea canopy and stems in frost protection, laying the foundation for further research on the energy distribution in tea ecosystem under the disturbance of airflow for frost protection. Assessing the Impact of Genotype-Specific Caprifig Fruit Storage on the Pollination Efficacy and Fruit Quality of “Bursa Siyahı” Cultivar: A Multivariate Analysis Approach

Fig types such as “Smyrna” and “San Pedro” require pollination (called caprification in fig cultivation) to produce a commercial crop, based on the crop and pollination characteristics of figs. Caprification is the process of hanging caprifig (male fig) fruits on female fig trees to ensure the transfer of pollen from the female fig to the caprifig by a wasp (Blastophaga psenes) that lives within the caprifig. It is necessary to extend the caprification period by using caprifig genotypes that ripen at different times, as female fig fruits ripen gradually. However, as caprifigs may not be continuously available for pollinating female figs, storing suitable caprifigs is necessary. The aim of this study was to assess changes in Blastophaga psenes, the duration of Blastophaga’s exit, and the viability of pollen from caprifigs of different genotypes (16 08 05, 16 08 09, 16 08 10, 16 09 10, and 16 ZF 08) stored for caprification. These stored caprifig genotypes were subsequently used for pollination three times at 8-day intervals, after which their impact on the set and quality of the edible fig fruits was evaluated. According to the average data, at the end of storage, the least B.psenes loss was obtained from the 16 08 05 (61.03%) genotype, and the highest was obtained from the 16 09 10 (67.00%) genotype. Pollen germination tended to increase with the storage of caprifig fruits, but this increase was not linear. After storage, the 16 08 09 and 16 09 10 genotypes exhibited greater pollen germination. The highest fruit set and quality were obtained when the 16 08 09 and 16 09 10 genotypes were used as pollen sources. Furthermore, since the 16 08 10 genotype is the latest ripening caprifig genotype, it has been determined that it can pollinate late-ripening “Bursa Siyahı” fruits. Principal component and path analysis demonstrated that pollen viability and germination rate were crucial in selecting caprifig genotypes for fruit set and quality. Full article

Berberis amurensis var. latifolia Nakai is a plant native to the Ulleung Island in Korea. In this study, we aimed to identify seed dormancy-breaking and germination requirements of this species using water imbibition experiments, gibberellic acid (GA3) treatment (0, 10, 100, or 1000 mg/L), cold stratification (0, 2, 4, 8, or 12 weeks at 5 °C), move-along experiments, and phenological studies. In the water imbibition experiment, the seed weight increased by more than 120% after 24 h. Analysis of the internal morphological characteristics of the seeds revealed that the embryo in freshly matured seeds was fully grown and did not grow thereafter. The final germination percentages after 12 weeks of cold stratification at 5 °C were 49 ± 6.4% and 63 ± 3.4% under light and dark conditions, respectively. In move-along and phenological studies, a longer cold stratification treatment period resulted in a higher germination percentage; however, the warm stratification treatment did not affect germination significantly. The GA3 treatment had little effect on seed germination. Therefore, we concluded that B. amurensis var. latifolia seeds have intermediate physiological dormancy, and pre-treatment with cold stratification for 12 weeks and incubation in the dark are required for effective seed propagation. Evaluation of Agronomic and Oil Characteristics of Selected Turkish Poppy Genotypes under Ankara’s Climate Conditions

Poppy is a minor agronomic field crop that is cultivated under a UN license. It is known for its alkaloids and seeds, and, rarely, for the latter’s use in ethnomedicine. Changing climate conditions could lead to the need for alternate areas for poppy cultivation in Türkiye. This experiment was conducted in Ankara, which is not a poppy production area. The morphological characteristics and oil characteristics of 19 Turkish poppy genotypes were determined over two years. According to the results, the emergence time was between 10 and 22 days, the flowering time ranged from 197 to 214 days, while the harvest maturation time was between 250 and 269 days. The plant height varied from 75.8 to 97.5 cm, the weight of 1000 seeds ranged from 305.0 to 428.0 mg, and the weight of the seeds per plant was between 2.95 and 5.78 g. Furthermore, the yield ranged from 100.7 to 202.3 kg da−1, the fat content was between 38.8 and 44.1%, and the protein content ranged from 15.9 to 18.4%. The linoleic acid content ranged from 66.77% to 75.60%, the oleic acid content ranged from 10.78% to 19.46%, and the palmitic acid content ranged from 8.38% to 9.90%. The highest yield in Ankara was obtained from the Çelikoğlu cultivar. Full article
