Epidermal melanin synthesis determines an individual’s skin color. In humans, melanin is formed by melanocytes within the epidermis. The process of melanin synthesis strongly depends on a range of cellular factors, including the fine-tuned interplay with reactive oxygen species (ROS). In this context, a role of cold atmospheric plasma (CAP) on melanin synthesis was proposed due to its tunable ROS generation. Herein, the argon-driven plasma jet kINPen® MED was employed, and its impact on melanin synthesis was evaluated by comparison with known stimulants such as the phosphodiesterase inhibitor IBMX and UV radiation. Different available model systems were employed, and the melanin content of both cultured human melanocytes (in vitro) and full-thickness human skin biopsies (in situ) were analyzed. A histochemical method detected melanin in skin tissue. Cellular melanin was measured by NIR autofluorescence using flow cytometry, and a highly sensitive HPLC-MS method was applied, which enabled the differentiation of eu- and pheomelanin by their degradation products. The melanin content in full-thickness human skin biopsies increased after repeated CAP exposure, while there were only minor effects in cultured melanocytes compared to UV radiation and IBMX treatment. Based on these findings, CAP does not appear to be a useful option for treating skin pigmentation disorders. On the other hand, the risk of hyperpigmentation as an adverse effect of CAP application for wound healing or other dermatological diseases seems to be neglectable. Full article

The aim of the presented research was to determine the suitability of both non-modified and modified buckwheat husk (BH) as a filler for urea–formaldehyde adhesive in plywood production. The effect of two modification methods, acetylation and silanization, was investigated. Infrared spectroscopy outcomes confirmed that both acetylation and silanization of the filler had occurred. Based on the results, it was found that the introduction of BH had a significant effect on both the adhesive properties and the characteristics of the manufactured plywood. The application of non-modified husks led to a reduction in viscosity and an extension of the gelation time, and the produced plywood boards were characterized by reduced bonding quality and increased delamination. Modification of the husk surface by acetylation and silanization with 3-aminopropyltriethoxysilane contributed to the noticeable improvement in the resin properties. On the other hand, the improvement in plywood properties, consisting of the increase in bonding quality and reduced delamination, was observed only in the case of the silanized husk. Furthermore, the use of non-modified and acetylated husk did not significantly influence the formaldehyde emission. The reduction in the investigated emission of formaldehyde was observed only in the case of variants containing 15 and 20% of silanized buckwheat husk. Full article

Learning to Read in an Intermediate Depth Orthography: The Longitudinal Role of Grapheme Sounding on Different Types of Reading Fluency

Phonological processing skills, such as phonological awareness, are known predictors of reading acquisition in alphabetic languages with varying degrees of orthographic complexity. However, the role of multi-letter-sound knowledge, an important foundation for early reading development, in supporting reading fluency development remains to be determined. This study examined whether two core foundational skills, phonemic awareness and grapheme sounding, have a predictive role in reading fluency development in an intermediate-depth orthography. The participants were 62 children learning to read in European Portuguese, and they were longitudinally assessed on phonemic awareness, complex grapheme sounding, and reading fluency (decoding, word, and text) from Grade 2 to Grade 3. The results showed that grapheme sounding predicted reading fluency development controlled for nonverbal intelligence and vocabulary, short-term verbal memory, and phonemic awareness. Grapheme sounding plays a prominent role in predicting reading fluency outcomes, whereas phonemic awareness (both accuracy and time per correct item) did not contribute to any of the three types of reading fluency. The fact that grapheme-sounding predicted reading fluency is likely due to complex grapheme-phoneme correspondences being required to achieve proficient reading. These findings provide insights into the cognitive processes underlying reading development in intermediate-depth orthographies and have implications for early literacy instruction. Full article

This study aimed to investigate the impact of an 8-week aerobic dance intervention on postural balance in children. Forty-one children, aged 9 to 11, were randomly assigned to either an aerobic dance group (ADG) or a control group (CG) from a primary school. Postural balance was assessed using center of pressure (CoP) excursions before and after the 8-week intervention period. Evaluations were conducted on both firm and foam surfaces in bipedal and unipedal stances under open-eyes (OE) and closed-eyes (CE) conditions, as well as on both medial–lateral (ML) and anterior–posterior (AP) surfaces in a bipedal stance under OE conditions. The ADG exhibited significantly decreased CoPVm values during firm bipedal CE, unipedal OE, foam bipedal OE and CE, and foam unipedal OE (p < 0.005). This study suggests that aerobic dance intervention improved postural balance in children, showcasing adaptability and improved stability under various conditions. Full article

Research has suggested that near future events are typically viewed from a first-person (an own-eyes, also known as field) perspective while distant future events are typically viewed from a third-person (an observer) perspective. We investigated whether these distinct mental perspectives would be accompanied by distinct eye movement activities. We invited participants to imagine near and distant future events while their eye movements (i.e., scan path) were recorded by eye-tracking glasses. Analysis demonstrated fewer but longer fixations for near future thinking than for distant future thinking. Analysis also demonstrated more “field” mental visual perspective responses for near than for distant future thinking. The long fixations during near future thinking may mirror a mental visual exploration involving processing of a more complex visual representation compared with distant future thinking. By demonstrating how near future thinking triggers both “field” responses and long fixations, our study demonstrates how the temporality of future thinking triggers both distinct mental imagery and eye movement patterns. The constant use of alcoholic beverages can deregulate serotonin levels, affecting neurotransmitters and triggering symptoms of anxiety. In this context, the objective of this work was to evaluate the anxiolytic potential and possible action mechanisms of the natural compound amentoflavone against the deleterious effects caused by alcohol withdrawal on the behavior of adult zebrafish (aZF). The experiments showed that amentoflavone did not change locomotion and did not cause toxicity in aZF during up to 96 h of analysis, with a median lethal concentration (LC50) greater than 1.0 mg/mL. The reversal of anxiety by pretreatment with granisetron suggested that the anxiolytic effect of amentoflavone is dependent on serotonergic 5-HT3A/3B receptors. Furthermore, amentoflavone reversed anxiety due to flumazenil pretreatment, suggesting a dependence on the GABAA receptor. The three concentrations of amentoflavone tested were effective in treating anxiety resulting from alcohol withdrawal. In silico analysis validated the in vivo results, supporting the idea that the interaction of amentoflavone with the protein occurs in a more stable manner than reference compounds. Amid growing interest in natural alternatives to treat anxiety disorders, amentoflavone is a potential candidate for a new anxiolytic compound that acts specifically on the 5HT3A/3B and GABAergic serotonergic pathways. A drastic level of resource degradation was revealed through a preliminary evaluation (completed in 2016) of water quality in a recreational lake in the second industrial city in Dammam, Saudi Arabia. The primary signs were a foul smell, algal bloom, high turbidity, and lack of aquatic life. This study aims to evaluate the influence of reverse osmosis (RO) on lake water quality. The recreational lake consists of two connected lakes (Lakes 1 and 2), which receive treated effluent from an industrial wastewater treatment plant. Composite samples were collected from the lakes to analyze their physiochemical parameters. Descriptive analyses were performed, and two water quality indices were developed to observe the variations in water quality conditions between the two periods (2016 and 2021). The results indicated that the water parameters of total dissolved solids (TDS), sulphate (SO42−), biological oxygen demand (BOD), and dissolved oxygen (DO) in 2016 (3356, 4100, 516, and 1.32 mg/L, respectively) were significantly improved in 2021 (2502, 1.28, 9.39, and 7.79 mg/L, respectively). The results of the water quality index (WQI) and comprehensive pollution index (CPI) indicated that the water quality in Lake 1 was significantly enhanced in 2021 (WQI = 85, CPI = 1) in comparison with assessment data from 2016 (WQI = 962, CPI = 8). However, the data from Lake 2 revealed higher pollution levels in 2021 (WQI = 1722, CPI = 18) than those recorded in 2016 (WQI = 1508, CPI = 13). As indicated by the absence of bad smells, algal blooms, and restoration of aquatic life, the RO intervention successfully improved the water quality in Lake 1. The WQI and CPI were helpful tools for evaluating lake water quality. Full article

This study focuses on the relationship between the outdoor environment and usage behavior of open spaces in cities with snowy and cold climates (winter cities), using an outdoor survey conducted at AKAPURA Plaza in Sapporo, Japan. This study seeks to understand walking and staying behaviors and analyzes their relationships to snowy outdoor environments. An analysis of the survey data shows that while the number of pedestrians using AKAPLA Plaza decreased as temperatures dropped, the ratio of staying behavior to the number of pedestrians did not decrease despite dropping temperatures. This study identified the following three design principles that can be applied to encourage the usage of open urban spaces during the winter. In winter, the number of pedestrians decreases in correlation with decreases in temperature; however, during the snowy season, walking can be encouraged by providing areas with less snow (PATH). Partially snow-covered areas can encourage photography and snow play behavior (STORAGE). Finally, providing walking routes to snow-covered areas can encourage staying behavior (APPROACH). These design principles were established based on the study in Sapporo and have the potential to be widely applied in other winter cities across the world through future research and analysis. A severe condition of sepsis can be a complication of necrotizing enterocolitis (NEC), which can occur in premature infants and becomes a medical challenge in the neonatal intensive care unit (NICU). It is a multifactorial intestinal disease (can affect both the small and large intestine) that can lead to ischemia of the intestinal tissues that evolves into acute organ necrosis. One of these factors is that different types of nutrition can influence the onset or the progression of the disease. Cow-milk-based infant formulas have been shown to cause it in premature infants more frequently than human milk. Recently, nutrition has been shown to be beneficial after surgery. Several issues still under study, such as the pathogenesis and the insufficient and often difficult therapeutic approach, as well as the lack of a common and effective prevention strategy, make this disease an enigma in daily clinical practice. Recent studies outlined the emerging role of the host immune system and resident gut microbiota, showing their close connection in NEC pathophysiology. In its initial stages, broad-spectrum antibiotics, bowel rest, and breastfeeding are currently used, as well as probiotics to help the development of the intestinal microbiota and its eubiosis. This paper aims to present the current knowledge and potential fields of research in NEC pathophysiology and therapeutic assessment. Full article

Highly contagious diseases, such as mumps, are a global concern as new epidemics continue to emerge, even in highly vaccinated populations. The risk of transmission and spread of these viruses is even higher for individuals who are more likely to be exposed, including healthcare workers (HCWs). In healthcare settings, both HCWs and patients are at risk of infection during the care process, potentially leading to nosocomial epidemic outbreaks. Mumps is often underestimated compared with measles and rubella, despite being milder and less likely to spread. In fact, the risk of complications following mumps infection is extremely high, especially if the disease occurs in adulthood. The measles–mumps–rubella (MMR) vaccine has been shown to be an excellent preventive measure. Unfortunately, the mumps component appears to be less effective in inducing immunity than those for measles and rubella (two-dose effectiveness of 85%, 95% and 97%, respectively). The main aim of our study was to investigate the prevalence of detectable mumps antibodies (serum IgG antibodies) in a cohort of Italian and foreign HCWs in relation to personal and occupational factors. We included in the study 468 subjects who underwent health surveillance at the Occupational Medicine Unit of the Tor Vergata Polyclinic in Rome during the period from January 2021 to March 2023. In our study, the proportion of HCWs found to be unprotected against mumps was very high (8.3%), and those found to be immune are below the WHO threshold for herd immunity (95%). From our data, it seems essential that all occupational health services carry out an accurate screening with a dose of anti-mumps antibodies to assess serological protection before starting a job, regardless of an individual’s vaccination history. This approach is proving to be beneficial, accurate, as it allows all serologically non-immune individuals to be vaccinated in the workplace, including those who would be protected by their vaccination history but have lost the antibody response. The Association of Central Sensitisation with Depression, Anxiety, and Somatic Symptoms: A Cross-Sectional Study of a Mental Health Outpatient Clinic in Japan

For patients with chronic pain and persistent physical symptoms, understanding the mechanism of central sensitisation may help in understanding how symptoms persist. This cross-sectional study investigated the association of central sensitisation with depression, anxiety, and somatic symptoms. Four hundred and fifteen adults attending an outpatient psychosomatic clinic were evaluated. Participants completed the Hospital Anxiety and Depression Scale, Somatic Symptom Scale 8, and the Central Sensitisation Inventory. The relationships between these factors were examined using descriptive statistics and multiple logistic regression analyses. The mean age was 42.3 years, and 59% were female. The disorders included adjustment disorders (n = 70), anxiety disorders (n = 63), depressive disorders (n = 103), feeding and eating disorders (n = 30), sleep–wake disorders (n = 37), somatic symptoms and related disorders (n = 84), and others (n = 28). In multiple logistic regression analyses, higher central sensitisation was associated with more severe anxiety, depression, and somatic symptoms after controlling for potential confounders. In the disease-specific analysis, somatic symptoms correlated more positively with central sensitisation than with depression or anxiety. Central sensitisation and depression, anxiety, and somatic symptoms were associated with patients attending an outpatient clinic. These findings highlight the importance of evaluating depression, anxiety, and somatic symptoms when assessing central sensitisation. Full article

Modelling size-realistic nanomaterials to analyse some of their properties, such as toxicity, solubility, or electronic structure, is a current challenge in computational and theoretical chemistry. The representation of the all-atom three-dimensional structure of a nanocompound would be ideal, as it could account explicitly for structural effects. However, the use of the whole structure is tedious due to the high data management and the structural complexity that accompanies the surface of the nanoparticle. Developing appropriate tools that enable a quantitative analysis of the structure, as well as the selection of regions of interest such as the core-shell, is a crucial step toward enabling the efficient analysis and processing of model nanostructures. The aim of this study was twofold. First, we defined the NanoFingerprint, which is a representation of a nanocompound in the form of a vector based on its 3D structure. The local relationship between atoms, i.e., their coordination within successive layers of neighbours, allows the characterisation of the local structure through the atom connectivity, maintaining the information of the three-dimensional structure but increasing the management ability. Second, we present a web server, called ATENA, to generate NanoFingerprints and other tools based on the 3D structure of the nanocompounds. A case study is reported to show the validity of our new fingerprint tool and the usefulness of our server. The scientific community and also private companies have a new tool based on a public web server for exploring the toxicity of nanocompounds. Full article

The aim of this study was to establish whether multiple blood parameters might predict an early treatment response to intravitreal bevacizumab injections in patients with diabetic macular edema (DME). Seventy-eight patients with non-proliferative diabetic retinopathy (NPDR) and DME were included. The treatment response was evaluated with central macular thickness decrease and best corrected visual acuity increase one month after the last bevacizumab injection. Parameters of interest were the neutrophil-to-lymphocyte ratio (NLR), monocyte-to-lymphocyte ratio (MLR), platelet-to-lymphocyte ratio (PLR), systemic immune-inflammation index (SII), vitamin D, and apolipoprotein B to A-I ratio (ApoB/ApoA-I). The NLR (2.03 ± 0.70 vs. 2.80 ± 1.08; p < 0.001), MLR (0.23 ± 0.06 vs. 0.28 ± 0.10; p = 0.011), PLR (107.4 ± 37.3 vs. 135.8 ± 58.0; p = 0.013), and SII (445.3 ± 166.3 vs. 675.3 ± 334.0; p < 0.001) were significantly different between responder and non-responder groups. Receiver operator characteristics analysis showed the NLR (AUC 0.778; 95% CI 0.669–0.864), PLR (AUC 0.628; 95% CI 0.511–0.735), MLR (AUC 0.653; 95% CI 0.536–0.757), and SII (AUC 0.709; 95% CI 0.595–0.806) could be predictors of response to bevacizumab in patients with DME and NPDR. Patients with severe NPDR had a significantly higher ApoB/ApoA-I ratio (0.70 (0.57–0.87) vs. 0.61 (0.49–0.72), p = 0.049) and lower vitamin D (52.45 (43.10–70.60) ng/mL vs. 40.05 (25.95–55.30) ng/mL, p = 0.025). Alterations in the NLR, PLR, MLR, and SII seem to provide prognostic information regarding the response to bevacizumab in patients with DME, whilst vitamin D deficiency and the ApoB/ApoA-I ratio could contribute to better staging. Rapid and Sensitive Detection of Verticillium dahliae from Soil Using LAMP-CRISPR/Cas12a Technology

Cotton Verticillium wilt is mainly caused by the fungus Verticillium dahliae, which threatens the production of cotton. Its pathogen can survive in the soil for several years in the form of microsclerotia, making it a destructive soil-borne disease. The accurate, sensitive, and rapid detection of V. dahliae from complex soil samples is of great significance for the early warning and management of cotton Verticillium wilt. In this study, we combined the loop-mediated isothermal amplification (LAMP) with CRISPR/Cas12a technology to develop an accurate, sensitive, and rapid detection method for V. dahliae. Initially, LAMP primers and CRISPR RNA (crRNA) were designed based on a specific DNA sequence of V. dahliae, which was validated using several closely related Verticillium spp. The lower detection limit of the LAMP-CRISPR/Cas12a combined with the fluorescent visualization detection system is approximately ~10 fg/μL genomic DNA per reaction. When combined with crude DNA-extraction methods, it is possible to detect as few as two microsclerotia per gram of soil, with the total detection process taking less than 90 min. Furthermore, to improve the method’s user and field friendliness, the field detection results were visualized using lateral flow strips (LFS). The LAMP-CRISPR/Cas12a-LFS system has a lower detection limit of ~1 fg/μL genomic DNA of the V. dahliae, and when combined with the field crude DNA-extraction method, it can detect as few as six microsclerotia per gram of soil, with the total detection process taking less than 2 h. In summary, this study expands the application of LAMP-CRISPR/Cas12a nucleic acid detection in V. dahliae and will contribute to the development of field-deployable diagnostic productions. The ClassMaps strategy builds resilience into classrooms by translating the compelling results of developmental risk and resilience research into simple action plans that embed positive protective supports into classroom routines and practices. The refined routines reinforce students’ developing self-agency and foster their nurturing relationships with adults and peers. The strategy is carried out in four stages—administering the ClassMaps Survey to describe students’ experiences of classroom strengths and weaknesses, conducting a classroom meeting with students to scrutinize the results and brainstorm plans for change, implementing simple modifications to classroom routines and practices based on that plan, and evaluating the impact of the modifications a few weeks later. The ClassMaps Survey’s central role in the strategy is justified by the reliability of its subscales and their relation to the key protective supports identified in resilience research. The ClassMaps strategy is a useful example of ‘giving psychology away’. Full article

Diffuse intrinsic pontine glioma (DIPG), now referred to as diffuse midline glioma (DMG), is a highly aggressive pediatric cancer primarily affecting children aged 4 to 9 years old. Despite the research and clinical trials conducted to identify a possible treatment for DIPG, no effective drug is currently available. These tumors often affect deep midline brain structures in young children, suggesting a connection to early brain development’s epigenetic regulation targets, possibly affecting neural progenitor functions and differentiation. The H3K27M mutation is a known DIPG trigger, but the exact mechanisms beyond epigenetic regulation remain unclear. After thoroughly examining the available literature, we found that over 85% of DIPG tumors contain a somatic missense mutation, K27M, in genes encoding histone H3.3 and H3.1, leading to abnormal gene expression that drives tumor growth and spread. This mutation impacts crucial brain development processes, including the epithelial–mesenchymal transition (EMT) pathway, and may explain differences between H3K27M and non-K27M pediatric gliomas. Effects on stem cells show increased proliferation and disrupted differentiation. The genomic organization of H3 gene family members in the developing brain has revealed variations in their expression patterns. All these observations suggest a need for global efforts to understand developmental origins and potential treatments. Full article

This paper investigates the estimated return rate and optimal order quantity under three cross-border e-commerce return logistics modes: direct mail (from predecessor), in situ destruction (new), and insurance (new). The estimated return rate under each model was analyzed and it was found that different modes have different thresholds in delivery lead time (the time retailers need to deliver goods to customers), and within which the estimated return rate increases as the delivery lead time increases. And a size comparison of the estimated return rates for the three models was conducted. A profit model was constructed based on the estimated return rate model, the optimal order quantity was calculated, and the effects of different factors (tax, postage, and delivery lead time etc.) on it were analyzed. For the insurance model, the effect of bearing the insurance ratio between retailers and consumers on the optimal order quantity was examined. The goal of this paper was to construct a model of the estimated return rate for the two new modes and to compare the estimated return rate of the three modes, which provides a reference for retailers to choose among the diversified return logistics modes and then make the best ordering strategy according to the influence of different factors on the optimal order quantity. Progressive Failure Mechanism of Shield Tunnel Face in Complex Urban Geological Environment

The construction of multiple tunnels across inland rivers has had a significant influence on the improvement of the transportation infrastructure. The technology for constructing tunnels is progressing towards the development of larger cross-sections, longer distances, and the ability to withstand high hydraulic pressure in complex hydrogeological conditions, including high-permeability strata. In order to ensure the face stability of shield tunnels under high hydraulic pressure that crosses a fault fracture zone, it is necessary to study the progressive failure mechanism of shield tunnel faces induced by high hydraulic pressure seepage. This paper employs finite element numerical simulation software to methodically examine the variation in the characteristics of the water seepage field, limiting support force, and face stability failure mode of shield tunnels passing through fault fracture zones with high hydraulic pressure under varying fault fracture width zones. The results show that the formation hydraulic gradient will progressively widen when the tunnel face is located within the undisturbed rock mass and is advanced towards the area of fault fracture. This will raise the likelihood of instability in the shield tunnel and progressively raise the limiting support force on the tunnel face. Moreover, as the tunnel face nears the region of fault fracture within the undisturbed rock mass, the damage range increases gradually. In addition, due to the increase in seepage force, the angle between the failure area and the horizontal plane becomes more and more gentle. On the contrary, as the tunnel’s face moves closer to the undisturbed rock mass from the region of the fault fracture, the damage range gradually decreases, and the dip angle between the damage area and the horizontal plane becomes steeper and steeper due to the decreasing seepage force in the process. The study findings presented in this work are highly significant, both theoretically and practically, for the design and management of safety. Full article

Sesame oil has been widely used in the daily diet due to its high nutritional value. Sesame oil is extracted at industrial scales and also in small scale by cold pressing at different temperatures. In this research, sesame oil was extracted by pressing at four temperatures, namely, 30 (control sample), 60, 90 and 120 °C, to evaluate its effects on the quality of extracted oils. Oil extraction yields were increased from 38 to 51% by increasing the pressing temperature. The highest amount of peroxide and acid values were related to the oil extracted at 120 °C. Tocopherols and total phenol content were reduced by the increasing the pressing temperature, and the highest amounts of these bioactive components were related to the control sample. The results of the fatty acids profile showed that the composition of oils extracted at different temperatures did not differ significantly (p > 0.05). The results of the present study give a clear picture about the effects of different pressing temperatures on the sesame oil quality and extraction yield, and can be useful in the extraction unit optimization. Hyperlipidemia, characterized by elevated cholesterol, lipids, and triglycerides in the bloodstream, is linked to hepatic oxidative damage. Doenjang, a traditional Korean condiment made from fermented soybeans, is known for its health benefits, yet its anti-hyperlipidemic effects remain understudied. Our study aimed to assess the hypolipidemic and hepatic protective effects of Doenjang on male ICR mice fed a high-fat cholesterol diet for 8 weeks. Mice were divided into three groups: the normal diet (ND), the high-fat cholesterol diet (HD), and the Doenjang-supplemented HD diet (DS) group. Doenjang supplementation significantly regulated total cholesterol, triglycerides, LDL cholesterol, and HDL cholesterol levels compared to the HD group. It also downregulated lipogenic genes, including PPARγ, FAS, and ACC, and positively influenced the cholesterol metabolism-related genes HMGCR and LXR. Moreover, Doenjang intake increased serum glutathione levels, activated oxidative stress defense genes (NRF2, SOD, GPx1, and CAT), positively modulated inflammation genes (NF-kB and IL6) in hepatic tissue, and reduced malondialdehyde levels. Our findings highlight the effectiveness of traditional Doenjang in preventing diet-induced hyperlipidemia and protecting against hepatic oxidative damage. Full article

With the rapid advancement of urbanization, the development and utilization of urban underground space resource (UUSR) has become one of the dominant features. However, in certain areas, the development of UUSR may cause disasters and accidents, such as ground collapse, settlements, and tunnel water gushing. Geological environmental factors (GEFs) are recognized as the fundamental constraining factor of UUSR development. In this paper, quality based on GEFs is defined to assess the development difficulty degree of UUSR. A 3D assessment framework is proposed based on 3D geological modelling and the interval continuous mathematical model (ICMM). The subjective and objective joint weight method of analytic hierarchy process and entropy weight method (AHP–EWM) is utilized to determine the weight of each indicator. The quality index (QI) of each spatial node of the 3D geological model is calculated by the ICMM mathematical model. A case study conducted in the Jiangbei New District of Nanjing, China, serves as a demonstration of the UUSR assessment. The results clearly illustrate the 3D distribution characteristics of the quality in the study area, offering valuable insights for future 3D urban underground space planning. Full article

Amyloid beta peptides (Aβ) have been identified as the main pathogenic agents in Alzheimer’s disease (AD). Soluble Aβ oligomers, rather than monomer or insoluble amyloid fibrils, show red blood cell (RBC) membrane-binding capacity and trigger several morphological and functional alterations in RBCs that can result in impaired oxygen transport and delivery. Since bioactive lipids have been recently proposed as potent protective agents against Aβ toxicity, we investigated the role of sphingosine-1-phosphate (S1P) in signaling pathways involved in the mechanism underlying ATP release in Ab-treated RBCs. In RBCs following different treatments, the ATP, 2,3 DPG and cAMP levels and caspase 3 activity were determined by spectrophotometric and immunoassay. S1P rescued the inhibition of ATP release from RBCs triggered by Ab, through a mechanism involving caspase-3 and restoring 2,3 DPG and cAMP levels within the cell. These findings reveal the molecular basis of S1P protection against Aβ in RBCs and suggest new therapeutic avenues in AD. Full article
